An alternative, and more lengthy procedure for the preparation of lysosomes follows folows the techniques of Sawant et al (Sawant, P.L., S. Shibko, U.S. Kumta and A.L. Tappel, 1964. BIOCHIM BIOPHYS ACTA 85,82.). This alternative will yield a purer lysosome fraction, but requires many more centrifugation steps. The procedure listed in this exercise takes advantage of the fact that lysosomes will aggregate in the presence of Ca&*SUPERSCRIPT ++. This procedure has the advantage of using only a preparative centrifuge, while reducing the number of centrifugation steps.
