1. Add the following to 3/4^th's the final volume of water:

2.0 grams/liter uracil minus drop-out base (USBiological D9536)
6.7 grams/liter yeast nitrogen base (USBiological Y2025)
20.0 grams/liter carbon source (2% w/v)*, either D(+)glucose (USBiological G3050) or D(+)galactose (USBiological G1030)

3. Sterilize media before use.

a. If the media is to become plates:

pour into a erlenmeyer flask with a capacity at least twice the volume of the media

label the flask

add 20 grams/liter of agar and cover the 'cap' the flask with tin foil

autoclave for 20 minutes on liquid cycle (program 1 on the autoclave)

cool > 30 minutes in a > 50^oC water bath

pour ~30 ml @ into sterile petri plates

label petri plates

b. If the media is to remain liquid:
• pour into the desired culture flask (2 liter erlenmeyer for 1 liter media) or into storage bottles
• DO NOT add agar but either 'cap' the flask with tin foil or place bottle caps LOOSELY on bottle top
• autoclave for 20 minutes on liquid cycle (program 1 on the autoclave)
• cool to desired temperature before use
